寒冷应激对小鼠脑内一氧化氮及一氧化氮合酶的影响研究

摘要:

目的:探讨寒冷应激(-10℃、10h)对小鼠脑内一氧化氮(NO)及一氧化氮合酶(NOS)的影响及意义。方法:将40只雄性昆明品系小鼠随机分为对照组和实验组,每组20只。寒冷应激实验结束后取脑组织检测一氧化氮含量及一氧化氮合酶活性。结果:对照组NO及NOS分别为21.12±8.68μmolgprot、2.21±0.57Umgprot。实验组为16.03±6.98μmolgprot、2.84±0.79Umgprot。寒冷应激后NO水平降低并有统计学意义(t=2.58,P<0.05),NOS活性增高并有统计学意义(t=-2.72,P<0.05)。应激后NO与NOS呈显著性负相关(r=-0.461,P<0.05)。结论:神经递质NO及NOS参与了中枢神经寒冷应激反应,且NO水平降低、NOS活性增高,可能对中枢神经细胞主要起保护作用。在寒冷应激后期吸入适量NO可能增强脑神经元对寒冷应激的耐受性。

Objective:To investigate the effect of cold stress (-10℃?10h) on NO and NOS in brain of mice.Methods:Forty mice were assigned into two groups randomly:The experimental group was the stress group and the other was the control group.The levels of NO and NOS activity were determined after cold stress.Results:The level of NO in stress group was significant lower than that in control group (t=2.67,P<0.05).NOS activity in stress group was higher than that in control group (t=-2.65,P<0.05).Correlation analysis showed that there was a negative relation between NO and NOS (r=-0.461,P<0.05).Conclusion:The increase of NOS and reduction of NO were possible factors that protected brain in cold stress.It may help to enhance the tolerance of brain to inhale NO in anaphase of cold stress.
